The proteolytic conversion of polynucleotide phosphorylase to a primer-dependent form.
نویسنده
چکیده
Polynucleotide phosphorylase from Micrococcus luteus (formerly Micrococcus lysodeikticw) can be converted from a primer-independent form (Form-I) to a primer-dependent form (Form-T) by limited proteolysis with trypsin. The conversion of Form-I to Form-T is accompanied by the breaking of between five and six peptide bonds per molecule of enzyme, and results in the removal of a small portion of the peptide chain of each subunit. The two enzymes have molecular weights of 2.6 X lo5 and 2.2 X 106, respectively, and both are composed of four subunits. Restoration of primer independence to Form-T is obtained by treatment of Form-T with sulfhydryl reagents. After thiol treatment, one or two enzyme sulfhydryl groups become titratable.

Aging or digestion with trypsin causes the enzyme to develop a primer requirement that is not eliminated by beta-mercaptoethanol. 3. Polynucleotide phosphorylase (polyribonucleotide:orthophosphate nucleotidyltransferase, EC 2.7.7.8) activity has been found in many prokaryotes and studied in detail since 1955. Such enzymes have been detected also in plants. We now describe the purification of polynucleotide phosphorylase from cucumber cotyledons and leaves. This enzyme is a complex of three subunits, possibly not identical, o...

Nicotinamide mononucleoside 5'-diphosphate in its reduced form is an excellent substrate for polynucleotide phosphorylase from Micrococcus luteus both in de novo polymerization reactions and in primer extension reactions. The oxidized form of the diphosphate is a much less efficient substrate; it can be used to extend primers but does not oligomerize in the absence of a primer. The cyanide addu...
متن کاملThe effect of trypsin digestion on the activities of polynucleotide phosphorylase.
1. Treatment of Micrococcus lysodeikticus polynucleotide phosphorylase (nucleoside diphosphate-polynucleotide nucleotidyltransferase) with trypsin causes a preferential loss of its cytidine diphosphate and uridine diphosphate polymerization activities. 2. The phosphorolytic activity of the enzyme towards polycytidylic acid is unaffected in conditions in which the cytidine diphosphate-polymeriza...

It is already known that modification of E. col i polynucleotide phosphorylase by endogenous proteolysis induces drastic changes in both phosphorolysis and polymerisation reactions. The structural parameters of the proteolysed polynucleotide phosphorylase are described. The phosphorolysis of polynucleotide, which is quite progressive for the native enzyme, is shown to be only partially progress...
